There are regular buses that stop directly outside the port gates that will take you into the main city.
Cadiz is just off the A4, which gives it great links from Madrid, Seville and Cordoba. If you are travelling from Barcelona and the North you can also use the N340. The Taglo train also runs twice a day from the centre of Madrid, giving a fast and efficient way of travelling to Cadiz.
